Because the MUC1 cytoplasmic tail domain (CTD) has many biological roles including tumor progression and cell adhesion disturbance and is expected to be expressed in isolated cancer cells, we raised a novel monoclonal antibody (MAb) MUC1-014E against an intracellular nonrepeating 19-amino-acid sequence (RYVPPSSTDRSPYEKVSAG: N-1217-1235-C) of the MUC1 CTD, using a synthetic peptide including the 7-amino-acid epitope (STDRSPY: N-1223-1229-C).
In the immunohistochemical staining of 107 gastrectomy specimens including 48 por2 and 31 sig lesions, the MAb MUC1-014E showed high rates of positive
staining (a parts per thousand yen5% of carcinoma cells stained) for por2 (100%) and sig (97%), and of the highest intensity staining (4+, a parts per thousand yen75% of carcinoma cells stained) for por2 (100%) and sig (90%). In the 89 biopsy PCI-34051 specimens including 82 por2 and 38 sig lesions, the MAb MUC1-014E showed high rates of positive staining for
por2 (100%) and sig (100%) and of 4+ staining for por2 (87%) and sig (84%). All the rates were significantly higher than those with cytokeratins (AE1/AE3 or CAM5.2).
The MAb MUC1-014E is very useful for accurate detection of isolated cancer cells in scirrhous gastric cancers.”
“Cynara scolymus (artichoke), Silybum marianum (milk thistle) and Cochlospermum angolensis (borututu) are three plants widely used regarding hepatoprotective effects but to the best of Z-VAD-FMK mechanism of action our knowledge no anti-hepatocellular carcinoma activity has been studied in the most consumed forms: infusions and dietary supplements. Herein, antioxidant properties, Selleck HSP990 anti-hepatocellular carcinoma activity and toxicity of infusions and dietary supplements of the mentioned plants were evaluated and compared. All the samples revealed antioxidant properties with EC50 values lower than the daily recommended dose, but infusions showed higher biological activity than dietary supplements. Borututu infusion gave the highest antioxidant activity (EC50 <= 170 mu g/mL), and also revealed anti-hepatocellular carcinoma activity (GI(50) = 146 mu g/mL) without toxicity in non-tumour liver cells (GI(50) > 400 mu g/mL). Artichoke infusion also presented antitumour
activity (GI(50) = 52 mu g/mL) but with toxicity for normal cells at a higher concentration (GI(50) = 72 mu g/mL). The antioxidant and antitumour properties were positively correlated with phenolics and flavonoids content. Overall, among the three studied species, borututu infusion proved to be the most complete sample regarding antioxidant and anti-hepatocellular carcinoma activity. (C) 2013 Elsevier B.V.
